From: Second harmonic generation light quantifies the ratio of type III to total (I + III) collagen in a bundle of collagen fiber

Two equations about SHG intensities are required to estimate λIII. Two relationships of type III ratio–SHG intensity (λIIIISHG-λIII) (a) and fiber bundle diameter–SHG intensity of type I collagen (DISHG-TypeI) (b) were obtained from separately performed experiments to determine k1, k2, and k3. For a tested collagen fiber bundle where λIII is unknown, D and SHG light intensity (ISHG) of the fiber bundle were measured (picture in the right bottom in (b). ISHG-TypeI was calculated from the D in the graph (b). Since the ratio of ISHG:ISHG-TypeI equals to the ISHG-λIII:k2, λIII can be determined. This study used the diameter of fiber bundle because the diameter should be proportional to the amount of collagen fiber bundle. This figure was created using Adobe Illustrator (Adobe, CS6).
