Seed-dependent phosphorylation of α-synuclein in pre-synaptic region of primary-cultured neurons. (a) Seed-dependent phosphorylation of α-synuclein (Phos-αS) in the pre-synaptic region. Primary-cultured cells grown in vitro for 14 days were treated with α-synuclein monomer (upper) or pre-formed fibrils (PFFs) (lower), and after one day, the cells were fixed and stained with anti-synapsin I (green) and anti-phosphorylated α-synuclein (red). Nuclei were stained with DAPI. Merged images are shown. (b) Seed-dependent phosphorylation of α-synuclein in axons. Primary-cultured cells grown in vitro for 14 days were treated with α-synuclein pre-formed fibrils (PFFs), and after 3 days (upper) or 7 days (lower), the cells were fixed and stained with anti-tau (green) and anti-phosphorylated α-synuclein (red). Nuclei were stained with DAPI. Merged images are shown. (c) Confocal laser microscopy analysis of cells treated with DMSO or drugs. Primary-cultured cells were treated with DMSO (left), CilD (center) or STLC (right) and stained with anti-phosphorylated α-synuclein (upper, green) and anti-synapsin I (lower, red).
