Relaxation of supercoiling by nicking does not affect chromatin binding of HMGB1. U2OS^2FP cells were treated with (a) 25, 50, 100, 200, 400 and 800 µM H₂O₂ for 20 min, or (b) with 10, 100 and 1,000 µg/ml bleomycin for 2 h, or (c) irradiated with 6, 12, 25 and 50 Gy X-ray, then the mobility of GFP-HMGB1 was measured by point FRAP. Graphs show one representative experiment of three repeats. The statistical test used is one-way ANOVA (α < 0.05).
