SFaN reduces SREBP precursor and mature form protein levels. (A,C) Huh-7 cells were sterol-depleted by incubation in medium D for 16 h. The cells were then shifted into medium D in the presence of the vehicle, 1 μg/ml 25-HC, or the indicated concentrations of SFaN (A) or SFeN (C). After incubation for 3 h, whole-cell extracts underwent immunoblotting (IB) with anti-SREBP-1 (2A4), anti-SREBP-2, or anti-β-actin antibodies. (B,D) Huh-7 cells were sterol-depleted by incubation in medium D for 16 h. The cells were then shifted into medium D in the presence of 100 μM SFaN (B) or SFeN (D). After incubation for the indicated periods, whole-cell extracts underwent immunoblotting (IB) with anti-SREBP-1 (2A4), anti-SREBP-2, or anti-β-actin antibodies.
