Annealing temperature has a small effect on the Cts of N1 and N2 using a pooled SARS-CoV-2 positive specimen spiked into water (blue and orange dotted lines, respectively) or for N1 when spiked into a pooled negative specimen (blue solid line), but annealing temperature has a large effect on the Cts of N2 when a pooled positive specimen is spiked into a pooled negative specimen (orange solid lines).
