The color in the microscopic images codes the average lifetime of the pixels. (a) is near to the coverslip, (b–d) each 0.5 µm further into the sample. The arrows over the phasor plots always point to the same position. Near the coverslip in (a), ATTO 647N labeled phalloidin produces a separate peak to the left in phasor plot that dwindles further into the sample due to the reduced amount of actin at higher sample depths. Note the movement of the Abberior STAR 635P contribution in the phasor plot (major peak) relative to the arrow.
