Assessing possible drug synergy between homoharringtonine and either doxorubicin (a-d) or paclitaxel (e–h). Cell viability (a, b, e, f) was measured after 4 days of drug treatment over a range of drug concentrations for HCC1806 (a, c, e, g) and HCC1806 + HA-CREB3L1 (b, d, f, h) cells. Homoharringtonine EC50 for HCC1806 = 29 nM; 1:3 serial dilutions (0–333 nM). Doxorubicin EC50 for HCC1806 = 13 nM; 1:3 serial dilutions (0–111 nM). Paclitaxel EC50 for HCC1806 = 1.2 nM; 1:3 serial dilutions (0–111 nM). A zero-interaction potency score (ZIP; c, d, g, h) was calculated by the SynergyFinder R-package for each data point to measure possible drug synergy; ZIP > 10 indicates synergy.
