Cytotoxic effects of EPAC1 Activators in HUVECs. HUVECs were treated with a concentration range of I942 (3.9–500 µM) for 24 h, followed by incubation with a mixture of Alamar Blue and CFDA-AM dyes for 30 min in the dark. Fluorescence was then measured at 532/590 nm (for Alamar Blue) and at 485/535 nm (for CFDA-AM) ex/em wavelengths. Subsequently, cells were incubated with Neutral Red dye for 1 h in the dark, followed by cell solubilization and fluorescence reading at 532/645 nm ex/em wavelengths. Curves were fitted to the data collected in all three assays to obtain EC50 values, which are plotted here as a bar graph. Data are presented as means ± SEM (n = 3).
