Fc-TL1A-AKALFF was Immune to DcR3-Based Inhibition. T cells were treated with 0.1 mg of immobilized anti-CD3 antibody alone or in the presence of exogenous DcR3 at 10 mg/mL. Either Fc-scTL1A (red) or Fc-scTL1A AKALFF (blue) was used to co-stimulate T cells. Anti-CD28 Ab was used as a co-stimulatory positive control. Anti-CD3-activated T cells without co-stimulatory signal was used as negative control. IFNg levels were measured by MSD assay. Stastical significance was determined using an unpaired T-test for each construct and data represents averages from three independent experiments using a single T cell donor.
