(A) Design of the crRNAs targeting conserved regions of LASV L and GPC genes. Overview of the LASV crRNA design strategy. Conserved regions of the L (370 bp) and GPC (211 bp or 283 bp) were selected and consensus sequences for lineage II or lineage IV were constructed (supplementary Table 2 and supplementary materials: lineage II target sequences and lineage IV target sequences). The degenerate spacer sequences of the crRNAs were designed by tiling the 28nt sequences across the consensus target sequences. (B) Structure of the Cas13a crRNA and target.
