Olaparib reduces HSC potential in BCR/ABL1-expressing cells. (a) The colony-forming activity of HSCs from wild-type (WT) or BCR/ABL1 transgenic mice (BATg) was analyzed in serial replating experiments. Cells were maintained on agar containing 5 μM olaparib. Colony numbers on each plate are shown in the bar graph. The average colony number from three independent experiments (mean ± SD) is shown. (b) BFB formation was assessed in bone marrow hematopoietic stem cells (Lin-c-kit+Sca1+CD105+) obtained from Parp+/+, Parp1-/-, Parp1+/+BATg/-, and Parp1-/-BATg/- mice.
