The hTERT plasmid was transfected into human normal breast cells (MCF-12F) to induce hTERT overexpression. Subsequently, various concentrations of exogenous IGFBP-3 were added. Exogenous IGFBP-3 decreased the levels of hTERT mRNA (A) and protein (B) in a dose-dependent manner. In hTERT overexpressed cells, IGFBP-3 decreased telomerase activity (C) and increased senescence-associated ß-galactosidase activity (D) in a dose-dependent manner. a. p < 0.05, vs. normal cells; b. p < 0.05, vs. hTERT overexpressed cells without IGFBP-3; c. p < 0.05, vs. hTERT overexpressed cells with 50 ng IGFBP-3. Multiple comparisons were performed using one-way ANOVA with Bonferroni post-hoc analysis. Each bar represents the mean ± SD of three independent experiments.
