Expression of desmosomal gene and proteins. (A) Expression of PKP2 JUP, DSG2, DSP and DSC2 in total RNA extracts of hiPSC-D and Primary cells from HC subjects and ACM patients cultured in growth medium. GAPDH was used as house-keeping gene and qRT-PCR data are presented as the genes threshold cycles (Ct) with respect to the housekeeping gene GAPDH (ΔCt) (n = 3 biological replicates; Two-way ANOVA and Tukey’s post-test). (B) Representative images of WB analysis of proteins extracted from hiPSC-D and Primary cells from HC and ACM individuals cultured in growth medium, hybridized with specific antibodies for desmosomal proteins (anti-DSC2, anti-DSP, anti-DSG2, anti-PKP2 and anti-PG).
