Intracellular Ca2+ measurements were performed on Human tracheal epithelial cells (HTEpC). Intracellular Ca2+ concentrations ([Ca2+]i) were recorded as Fura-2/AM (Fura-2) fluorescence intensity ratio of 340:380 nm excitation. (A) When FURA-2 340/380 ratio was normalized after a 100-s resting period, fluorescence ratio remained constant under resting conditions during the 800-s observation period, demonstrating no changes in [Ca2]i. Arrows indicate different timepoints of the FURA-2 340/380 ratio, which are illustrated in (B1B3) after conversion into false colors. Potassium chloride (K+, 200 mM) completely inverted the membrane potential, leading to a strong increase in FURA-2 340/380 fluorescence of human tracheal epithelial cells.
