A Virotrap-based assay identifies calpain small subunit 1 (CAPNS1) as a possible TGM1 interactor. (a) Virotrap principle. A protein consisting of HIV-1 p55 Gag protein fused to the bait (TGM1, eDHFR as a negative control) is transiently expressed in HEK293T cells. Gag directs this fusion protein to the cell membrane and induces budding of virus-like particles (VLPs) in which interaction partners of the bait protein are trapped. The released VLPs are purified and the proteins are identified by mass spectrometry. (b) Volcano plot showing TGM1 interactors identified by Virotrap-based screening.
