Single cell sequencing data corroborates the upregulation of mtDNA-encoded genes in SCC CAFs. (A) UMAP analysis of single-cell sequencing data collected by Ji et al.27 from primary SCC CAFs and matched NFs from twelve donors. Eleven clusters were identified based on the expression of the gene markers listed in the legend. The fibroblasts cluster, accounting for ~ 5% of the sequenced cells, was analyzed further. (B) Analysis of the expression of conventional CAF biomarkers (grey bars, genes indicated in the legend) and mtDNA-encoded genes (green bars, genes indicated in the legend) in NFs and SCC CAFs from six patients with sufficient fibroblasts sequenced. NFs were set to 1 and relative expression fold changes in Log2 scale were calculated for each NFs and CAF pair (the asterisk indicates a comparison surviving Benjamini–Hochberg False Discovery Rate correction).
