Maize ZmCYP81A9 is inhibited by P450 probes. (a) Reaction scheme of hydroxylation of bentazone, catalyzed by ZmCYP81A9. (b) P450 probes inhibit ZmCYP81A9 activity. Microsomes isolated from yeast expressing ZmCYP81A9 were preincubated with 300 μM or without inhibitor and with or without 1 mM NADPH for 20 min at 28 °C, and then incubated for 120 min with 200 μM bentazone at 28 °C. Metabolites were extracted with acetonitrile and analyzed by LC–MS to measure peak areas of hydroxylated bentazone. The no-inhibitor-control was set at 100% activity and used to calculate the relative activity and statistical relevance of the other samples (t-test; ***P < 0.001). Error bars represent standard deviation of n = 3 replicates.
