Transiently expressed Maize ZmCYP81A9 is labeled by P450 probes. (a) Transient expression of GFP-tagged ZmCYP81A9 by agroinfiltration of Nicotiana benthamiana. (b) ZmCYP81A9-GFP localizes to the ER. ZmCYP81A9-GFP was transiently co-expressed with ER marker RFP-HDEL by agroinfiltration. Confocal images were taken 5 days after agroinfiltration. Scalebar 5 μm. Experiment was repeated twice with similar results. (c) ZmCYP81A9-GFP is labeled by the P450 probes. Microsomes were isolated from agroinfiltrated leaves expressing ZmCYP81A9-GFP and labeled for 1.5 h with various probes (10 μM) with or without 1 mM NADPH. Labeled proteins were coupled to Cy5-picolyl-azide via click chemistry and visualized by in-gel fluorescence scanning. Western blot with anti-GFP antibodies shows accumulation of ZmCYP81A9-GFP at its expected MW. *RBCL.
