Proposed model of LIN28B repression by SOX6. In fetal cells and in cancers with illegitimate LIN28B reactivation, LIN28B blocks the processing of Let-7 precursors to Let-7 miRNA. Let-7 reduction leads to the failure of silencing of proproliferative genes, including c-MYC and IGF2BP1, thus promoting cell proliferation. SOX6, by repressing LIN28B transcription, allows the maturation of Let-7 miRNAs, among the most abundant miRNAs in adult cells17.
