The Akt pathway is necessary for SAA to induce GSK-3β phosphorylation and Nrf2 nuclear translocation in primary cortical neurons. These effects were inhibited by treatment with SH-6 (an AKT inhibitor) and ML385 (an Nrf2 inhibitor). (A) Western blot analysis was used to detect the expression of phosphorylated and total Akt. Phosphorylated and total GSK-3β, Nrf2 proteins, GPX4 proteins, and XCT proteins. (BH) Western blot analysis was used to detect the expression of p-AKT, GSK-3β, Nrf2, GPX4, and XCT proteins after treatment with the inhibitor of the Akt/GSK-3β/ Nrf2 pathway. Values are expressed as means ± SEM.
