VDAC3 knockdown reversed the effect on cellular senescence and osteogenic differentiation in etoposide-induced BMSCs with ALKBH5 knockdown. qPCR was performed to measure the expression of (A) ALKBH5 and (B) VDAC3 in BMSCs following sh-ALKBH5 and sh-VDAC3 transfection. (C and D) Cellular senescence of BMSCs was assessed using a senescence β-galactosidase staining kit. The expression of (E) p21 and (F) p16 in BMSCs transfected with different plasmids was measured by qPCR. (G) The protein levels of RUNX2, COL1A1 and ALP in BMSCs transfected with different plasmids were measured by western blot. (H and I) ALP staining was conducted using a BCIP/NBT alkaline phosphatase colour development kit after 14 d of osteogenic differentiation induction on BMSCs. (J and K) An ARS staining kit for osteogenesis was performed to ARS staining after 14 d of osteogenic differentiation induction on BMSCs.
