Interfering with USP50 inhibits LPS-induced pyroptosis of macrophages. (A) THP-1 cells were treated with PMA to obtain macrophage phenotype. These macrophages were transfected with si-USP50 and si-NC, and the mRNA and protein levels of USP50 was measured by qPCR and immunoblotting, respectively. (B) Flow cytometry was performed to determine cell pyroptosis, and (C) pyroptosis rate was quantified. (D) IL-1β and (E) IL-18 levels in the supernatant of the cell culture medium were detected using ELISA. (F) The levels of pyroptosis-related proteins including ASC, NLRP3, GSDMD, GSDMD-N, pro-caspase-1, and cleaved caspase-1 were detected using immunoblotting. (n = 3).
