M1 macrophage-derived medium reduces MGMT and MPG expression in GBM cells by inhibiting STAT3 activation. (A) Effect of M1 macrophage medium (24-hour incubation) on STAT1/STAT3 activation and DNA repair enzymes (MGMT, MPG) was evaluated by Western blot. (B) Protein levels of DNA repair enzymes and transcription factors were analyzed in GBM cells transiently transfected with STAT1/STAT3 overexpression or empty vectors. (C) MGMT, MPG, p-STAT3, and STAT3 expression were analyzed in T98G cells treated with a STAT3 activation inhibitor. (D) Western blot analysis of MGMT and MPG in T98G cells transiently transfected with STAT3 or empty vectors, followed by treatment with conditioned medium from M1-differentiated or M0 control macrophages.
