(C) Western blot analysis of MADD expression in control and knockout cells after 24 h of treatment with 4-Hydroxytamoxifen (4HT). β-actin served as a loading control. (D) Immunocytochemistry of MADD expression. Cells were cultured on sterile coverslips in the 8-well chamber, and following fixation, the cells were stained with anti-MADD-specific antibodies. DAPI staining was used for counter-staining. (Green = MADD, blue = nucleus) absence of any green color indicates that MADD was successfully knocked out in ATC cells at the translational level.
