Tagging transmembrane protein Dpr12 in MB compartments using the dFlpTag system. Constitutive and conditional GFSTF and smGFP::HA tagging strains were generated by swapping Dpr12MI01695. (A) GFP-tagged Dpr12 was preferentially detected in MB γ4/γ5 compartments in both pupal and adult brains of constitutive tagging strains. (B) With different GAL4 drivers to express UAS-FLP, the corresponding cells were labeled by mtdTomato in Dpr12dFlpTag − GFSTF(cond.) fly brains. However, the GFP-tagged Dpr12 was only detected with R57C10-GAL4 and R13F02-GAL4, but not with R58E02-GAL4, which expresses in γ4/γ5-projecting dopamine neurons. Scale bars: 20 μm.
