Doxorubizen inhibits Topo II activity. The pHOT plasmid was incubated in the presence of DMSO (solvent control), doxorubicin, or doxorubizen (0.05, 0.1, 1, or 10 µM for both drugs) for 30 min at 37 °C. Doxorubizen (1 and 10 µM) prevented the formation of relaxed topoisomers, and the linearized pHOT DNA product accumulated.
