In the absence of substrates, MacA binds TolC to form the MacA-TolC subcomplex and then recruits MacB to assemble into the MacAB-TolC efflux pump. In the presence of substrates, MacAB-TolC hydrolyzes ATP to efflux substrates, and MacB dissociates from MacA-TolC subcomplex.
