a hTSCs were first transduced with a lentivector expressing Cas9 under the control of doxycycline, then with a lentivirus expressing two sgRNAs flanking the ATAC-11 element that was previously used to delete this element in BeWo cells. After selection with puromycin, polyclonal cells (dA11) were analyzed by RT-qPCR for selected ISGs and compared to control cells (Cas9). 10 µg/ml poly(I:C) was added overnight.
