a, HEK293T cells transiently transfected with IRF1-SFB for 24 h were treated with DMSO, 5 μM TSA, 5 mM NAM, or both compounds for another 24 h. Then, cell lysates were immunoprecipitated with streptavidin beads and immunoblotted with the indicated antibodies. b, The seven acetylated lysines identified by mass spectrometry were as follows: K43, K66, K70, K78, K117, K275, K299. c, HEK293T cells transiently cotransfected with vector or HA-KAT8 and the indicated mutants of IRF1-SFB for 48 h. Then, cell lysates were immunoprecipitated with streptavidin beads and immunoblotted with the indicated antibodies. d, HEK293T cells transiently cotransfected with IRF1 WT or K78R mutant and the indicated acetyltransferases for 48 h. Then, cell lysates were immunoprecipitated with streptavidin beads and immunoblotted with the indicated antibodies. e, HEK293T cells transiently cotransfected with IRF1-SFB and HA-KAT8 or HA-KAT8-C316S mutant as indicated for 48 h.
