ITK-SYK-expressing T cells were sorted by FACS for eGFP+ from the spleens of acutely induced ITK-SYKCD4-creERT2 (‘Pdcd1+/+) and ITK-SYKCD4-creERT2;Pdcd1−/− mice (‘Pdcd1−/−) and cultured in vitro for four hours. Next, the cell culture supernatants were harvested, and glucose concentrations were determined by colorimetric measurements. All animals received single-dose intraperitoneal injections of tamoxifen five days before the experiment (n = 6 and n = 4 mice per genotype). Data was normalized to viable cell number.
