a-d, Identification of tumor antigen-specific TCRVβ clonotypes by FEST assay on activated T cell expansion for PT05 (a), PT07 (b), PT14 (c) and PT16 (d). TIL (PT05 and PT16) or peripheral T cells (PT07 and PT14) were co-cultured with APCs loading neoantigen peptides. After 10 days of co-culture, TIL expansion was enriched and sequenced. Compared to “T cells + APCs” controls, expansion fold for each TCR clonotype was shown at each condition. CD4 or CD8 origin of each clonotype was labeled. ND: not detectable in CDR3Vβ sequencing data of flow sorted CD4+ or CD8+ TIL.
