a In vitro translation of TiLV segments 1–10. Full-length coding sequences of each segment were transcribed and translated in rabbit reticulocyte lysate supplemented with [³⁵S]-methionine/cysteine. Lysates were resolved by SDS-PAGE and translation products visualised by autoradiography. arrowheads indicate the predominant protein products. An additional independent experiment (lane 12, separate box) confirmed slow migration of the S10 product. b Expression of GFP-tagged TiLV proteins in 293 T cells. Cells were transfected with constructs encoding each polypeptide (S1–S10) fused to GFP, or with empty vectors. 48 h later, cell lysates were subjected to SDS-PAGE and immunoblotting using anti-GFP and anti-α-tubulin (loading control) antibodies.
