From: Single-cell transcriptome of early embryos and cultured embryonic stem cells of cynomolgus monkeys
The overall experimental design and work flow. The isolated cells dissociated from each samples were picked up for the cDNA synthesis. Then the quality and key gene expression of the generated cDNAs were examined by qPCR and the selected samples were used for the library construction. The libraries were sequenced and the resultant reads were mapped onto the cynomolgus monkey genome. The global distribution of gene expression in each cell was validated. Finally, the annotations of cell clusters were defined by comparing the gene expression patterns of the transcriptome and histological analysis such as immunofluorescent analysis.
