MafK-knockdown mice exhibit defective NF-κB signalling.
(a) Effects of MafK depletion on mRNA levels in the liver. Mice were transfected with the indicated MafK siRNAs for 72 h and liver samples were analysed by real-time PCR. (b) MafK-deficient mice were resistant to endotoxic shock. The mice were sensitized with GalN (700 mg/kg) followed by LPS (50 μg/kg) challenge and monitored hourly. The survival curve is presented as the percentage of survivors over time post-challenge. (c) MafK-knockdown mice are defective in NF-κB-dependent gene expression. MafK-knockdown mice were sensitized with LPS (15 mg/kg) challenge and gene expression was analysed. The indicated genes were measured by real-time PCR of liver RNA. Values are representative of at least three independent experiments. (d) MafK-knockdown mice were sensitized with LPS (15 mg/kg) challenge and cytokine expression was analysed. The indicated cytokines in blood were measured by ELISA.
