(A) Quantification of unprocessed RNA for four muscle-specific genes in C2C12 myoblasts cultivated in growth medium (GM) or differentiation medium (DM + I) and with control (CTL) siRNA or Mtor knockdown. mRNA was quantified by qRT-PCR, normalized to Gapdh and shown relative to expression in GM. (B) Quantitation of luciferase in C2C12 cells cultivated as above, following transfection with reporter plasmids containing the Muscle Creatine Kinase promoter (Ckm) or a minimal SV40 promoter driven by four MyoD-specific “E” boxes (4 R-SV).
