HL-60 cells were treated with indicated doses of NMHC for 24 h and the full length (FL) or cleaved NOTCH1 (ICN1) was detected by immunoblotting. Protein band intensity was quantified by ImageJ and ratios of ICN1/FL were calculated and plotted on the right. P-values were derived from Student’s t-test (*P < 0.05, **P < 0.01). The X-ray crystal structures of the NRR domain was from PDB (code: 3ETO) and molecular modeling method was employed to investigate the interaction of NMHC with NRR.
