The intensity of the punctate spots was abolished dramatically after the addition of proteinase K, which indicates that the majority of ClyA-GFP face up toward the bulk solution and that the orientation of the SB is the same as the OMV and bacterial cell. The bar graph below summarizes the densities of bright spots in the images. Multiple runs (n > 3) were performed to obtain bilayer diffusivities and particle numbers.
