Effect of exposure to SDS on structure and function of IgG.

The IgG fraction purified from the sera of rabbit immunized with α-LA was exposed to various concentrations of SDS, dialyzed and antigen binding activity assayed by ELISA (Panel a). Panel b shows the effect of 0.1% SDS on the antigen binding activity of anti-HRP IgG raised in rabbits and goats. Results are expressed as mean ± SD of three independent experiments. Rabbit IgG were incubated with various concentrations of SDS (detailed in Panel e also for Panel c,d) for 1 h, dialysed extensively against PBS, pH 7.4 and subjected to far-UV CD (Panel c), near-UV CD (Panel d) and intrinsic fluorescence (Panel e) spectral analysis. Results are expressed as mean of three independent experiments.
