Building autopoietic biochemical systems requires in situ tRNA synthesis, which remains challenging. Here, authors achieve continuous production of all 21 tRNAs in a PURE system using optimized DNA templates, enabling steady-state protein expression and advancing towards self-regenerating synthetic cells.
