Developmental signals control chromosome segregation fidelity during pluripotency and neurogenesis by modulating replicative stress

Here the authors show that the patterning signals WNT, BMP, and FGF control chromosome segregation fidelity during early lineage specification and neurogenesis, which could provide a rationale for the spatio-temporal distribution of genomic mosaicism during human development.

Replication-IDentifier links epigenetic and metabolic pathways to the replication stress response

Cells activate various pathways to counter DNA replication stress. Using Replication-IDentifier (Repli-ID), the authors identified 423 genes, including LGE1 and ROX1, linking epigenetic and metabolic pathways to the replication stress response.

Genome instability due to ribonucleotide incorporation into DNA

Polymerase exclusion of ribonucleotides during DNA replication is imperfect. New data indicate that DNA polymerase ϵ incorporates into DNA ribonucleotides that are repaired by an RNase H2–dependent process and that defective repair results in replicative stress and genome instability.

Evidence that processing of ribonucleotides in DNA by topoisomerase 1 is leading-strand specific

In the absence of RNase H2, ribonucleotides incorporated during DNA replication can be processed by Top1. This activity is directed to the nascent leading strand, because gaps in the lagging strand would limit torsional tension.
