The deubiquitinase A20 is a potent inhibitor of NF-κB signaling pathways. Ma and colleagues identify distinct roles for A20 ubiquitin-binding ZF4 and ZF7 domains, which exhibit different phenotypes upon mutation, but play synergistic roles in regulating inflammatory responses.

It is shown that ABIN-1 (a protein that interacts with the ubiquitin-editing enzyme A20 and is thought to negatively influence NF-κB signalling) is essential for life; without it, mice die during embryogenesis with fetal liver apoptosis and anaemia. This appears unrelated to the role of ABIN-1 in NF-κB signalling.

A20 is a deubiquitinating enzyme that restrict inflammation by various mechanisms. Ma and colleagues show that A20 inhibits necroptosis by inhibiting the ubiquitination of RIPK3 and formation of the RIPK1-RIPK3 complex.

The deubiquitinase A20 is a negative regulator of transcription factor NF-κB signaling pathways. Ma and colleagues show that mice lacking A20 expression in dendritic cells have defective lymphocyte homeostasis and develop spontaneous inflammatory disease.
