Several proteins have recently been found to consist of repetitive structural units in a superhelical arrangement. The structure of the leucine-rich repeat variant protein represents an unexpected addition to the list of such proteins, with the repetitive unit consisting of antiparallel α- and 3₁₀-helices.

An enzymatic dual-oxa Diels–Alder reaction constructs the oxygen-bridged tricyclic acetal unit of (–)-anthrabenzoxocinone

The hetero-Diels–Alder reaction is a powerful strategy for constructing six-membered heterocycles, yet natural enzymatic hetero-Diels–Alder reactions are limited to a single heteroatom. Now a bifunctional enzyme, Abx₍₋₎F, is found to catalyse the dehydration and a dual-oxa Diels–Alder reaction to form the oxygen-bridged tricyclic acetal of (−)-anthrabenzoxocinones.

MyD88 TIR domain higher-order assembly interactions revealed by microcrystal electron diffraction and serial femtosecond crystallography

MAL and MyD88 are downstream adaptors of Toll-like receptors (TLR) and the MAL TIR domain forms filaments in vitro, which in turn nucleate the assembly of crystalline arrays of the MyD88 TIR domain. Here, the authors present the structure of these MyD88 TIR crystalline arrays solved by both microcrystal electron diffraction and serial femtosecond crystallography, and they show with mutagenesis experiments that MyD88 interface residues are important for TLR4 signaling in vivo.

Free-electron lasers are capable of high repetition rates and it is assumed that protein crystals often do not survive the first X-ray pulse. Here the authors address these issues with a demonstration of multi-hit serial crystallography in which multiple FEL pulses interact with the sample without destroying it.

The PsaA binding protein delivers Mn²⁺ to the human pathogen Streptococcus pneumoniae. Structural and biochemical studies now explain its metal specificity, showing that metal binding induces a closed complex that is reversible for the desired substrate but irreversible for the inhibitor Zn²⁺.

Dysregulation of transition metal ion homeostasis is the molecular basis for cadmium toxicity in Streptococcus pneumoniae

The molecular basis for the high toxicity of cadmium is unclear. Here, Begg et al. use the bacterium Streptococcus pneumoniaeas a model system, and show that cadmium uptake increases sensitivity to oxidative stress by reducing intracellular concentrations of manganese and zinc through different mechanisms.

The new European X-Ray Free-Electron Laser (EuXFEL) is the first XFEL that generates X-ray pulses with a megahertz inter-pulse spacing. Here the authors demonstrate that high-quality and damage-free protein structures can be obtained with the currently available 1.1 MHz repetition rate pulses using lysozyme as a test case and furthermore present a β-lactamase structure.

Nicholas Hayward and colleagues sequenced eight metastatic melanoma exomes and identified frequent somatic mutations in two MAP kinase family genes, MAP3K5 and MAP3K9. Mutation in MAP3K9 may confer resistance to temozolomide, a common chemotherapeutic drug.
