A virus-encoded type I interferon decoy receptor enables evasion of host immunity through cell-surface binding

Secreted cytokine decoy receptors encoded by viruses can act as potent immune evasion proteins modulating antiviral immunity. Here Hernaez et al. show that cell surface binding is required for efficient evasion of the host response by a secreted virus encoded type I IFN decoy receptor of vaccinia and ectromelia virus using an in vivo model of infection.

SARS-CoV2-mediated suppression of NRF2-signaling reveals potent antiviral and anti-inflammatory activity of 4-octyl-itaconate and dimethyl fumarate

Viral infections usually cause disease through direct cytopathogenic effects and excessive inflammatory responses. Here, Olagnier et al. show that two NRF2 agonists, 4-OI and DMF, possess broad IFN-independent antiviral activity and decrease host inflammatory response to SARS-CoV-2 infection.

Enhanced virulence of mpox virus clade Ib over clade IIb in the CAST/EiJ mouse model

Monkeypox clade Ib virus shows enhanced spread, increased lung pathology, and systemic dissemination compared with clade IIb in CAST mice, revealing higher virulence and underscoring the need for surveillance of emerging lineages.

Chemokines cooperate with TNF to provide protective anti-viral immunity and to enhance inflammation

Cytokines play critical roles in the anti-viral response but their contribution in vivo remains unclear. Here the authors show that viral CrmD is a major virulence determinant and requires the SECRET and TNF binding domains to counter both the chemokine and cytokine driven inflammatory defenses.
