Stable human regulatory T cells switch to glycolysis following TNF receptor 2 costimulation

After activation, conventional T cells undergo metabolic reprogramming. de Kivit et al. show that in human thymic regulatory T cells, TNFR2 stimulation promotes a glycolytic switch with a preferential glucose-derived carbon flux into the TCA cycle to support suppressive functions.

P53-dependent hypusination of eIF5A affects mitochondrial translation and senescence immune surveillance

Here the authors discovered that senescent cells increase protein synthesis through eIF5A regulation, involving polyamine metabolism and p53 pathways, crucial for immune surveillance and mitochondrial protein production in aging and cancer contexts.

Fatty acid metabolism in aggressive B-cell lymphoma is inhibited by tetraspanin CD37

Tetraspanin CD37 deficiency has been reported as a prognostic marker for aggressive B-cell lymphoma. Here, the authors show that CD37 interacts with the fatty acid transporter 1 to inhibit palmitate uptake and its deficiency leads to increased fatty acid metabolism which promotes tumorigenesis in B-cell lymphoma.

Serine metabolism remodeling after platinum-based chemotherapy identifies vulnerabilities in a subgroup of resistant ovarian cancers

Metabolic reprogramming is associated with cancer development and therapy resistance. Here, the authors show that downregulation of the serine biosynthesis enzyme PHGDH in a fraction of patients is associated with relapse in platinum-treated ovarian cancers and to NAD⁺ and PARP activity upregulation.

The authors show that p53 helps cancer cells survive serine depletion by coordinating metabolic remodelling; a diet lacking serine slowed tumour growth in mice, with p53-null tumours showing greatest sensitivity to serine starvation.
