Probiotic therapy for colorectal cancer is limited by the poor tumor targeting and low intratumoral activity. The authors show that manganese-engineered L. reuteri with glycerol (MnLR/Gly) accumulates in colorectal tumors, produces cancer-selective metabolites, activates antitumor immunity, and achieves potent, durable tumor suppression in preclinical models.
