JHDM1B/FBXL10 is a nucleolar protein that represses transcription of ribosomal RNA genes

The histone demethylase JHDM1B represses expression of the ribosomal RNA genes by demethylating the trimethylated lysine 4 residue of histone H3. This effect on rRNA expression is correlated with proliferation defects, and so reduced levels of JHDM1B may contribute to tumour development.

Control of chromosome stability by the β-TrCP–REST–Mad2 axis

The transcription factor REST has been implicated in tumourigenesis, and β-TrCP regulates REST degradation. REST, in turn, controls the chromosome stability by regulating the expression of spindle checkpoint protein Mad2, suggesting that this mechanism contributes to the roles of β-TrCP and REST in transformation.

ERAP1 promotes Hedgehog-dependent tumorigenesis by controlling USP47-mediated degradation of βTrCP

ERAP1 is an endoplasmic reticulum aminopeptidase that trims MHC Class-I peptides for antigen presentation. Here, the authors show that ERAP1 enhances Hedgehog signalling by sequestering USP47 from βTrCP and promoting tumorigenesis through βTrCP degradation and increased Gli protein stability.

Itch/β-arrestin2-dependent non-proteolytic ubiquitylation of SuFu controls Hedgehog signalling and medulloblastoma tumorigenesis

SuFu is a tumour suppressor in medulloblastoma and regulates Gli proteins in the Sonic Hedgehog pathway; however, the molecular mechanisms behind this regulation are unclear. Here, the authors show that the Itch/β-arrestin2 complex binds and ubiquitylates SuFu, facilitating the interaction with Gli3 and its conversion into the repressive form, thus counteracting medulloblastoma formation.
