Niche-based screening identifies small-molecule inhibitors of leukemia stem cells

High-throughput screening systems that better mimic the physiological complexity of diseased tissues may aid the discovery of more efficacious compounds. A co-culture system that mimics the microenvironment of leukemia stem cells (LSCs) in bone marrow enables the discovery of compounds, including lovastatin, that selectively kill LSCs.

Mesenchymal and haematopoietic stem cells form a unique bone marrow niche

The identity of the cells that form the haematopoietic stem cell (HSC) niche in bone marrow has been unclear. These authors identify nestin-expressing mesenchymal stem cells as niche-forming cells. These nestin-expressing cells show a close physical association with HSCs and express high levels of genes involved in HSC maintenance, and their depletion reduces bone marrow homing of haematopoietic progenitors.

Hematopoietic stem cells (HSCs) migrate from the fetal liver to the bone marrow (BM) during embryogenesis. Here the authors show that the WAVE2 complex scaffold Hem1 is required for engraftment of HSCs in BM, not through its canonical role regulating actin polymerization, but through c-Abl survival signaling.

Live-animal tracking of individual haematopoietic stem/progenitor cells in their niche

High resolution combined confocal and two-photon video imaging of individual haematopoietic cells is performed in the bone marrow of living animals, examining their relationship to blood vessels, osteoblasts and endosteal surface as they home and engraft. It is found that osteoblasts were enmeshed in microvessals and different populations of haematopoeitic cells were localized in different areas according to their stage of differentiation. In settings of engraftment as well as expansion, marrow stem/progenitor cells were in closer proximity to bone and osteoblasts.

Myeloproliferative neoplasms are diseases that arise in the stem cells of the blood. In a recent paper published in Nature, Arranz et al. demonstrated that abrogation of sympathetic nerve fibers reduced bone marrow Nestin⁺ mesenchymal cells, which in turn led to an expansion of hematopoietic stem cells and progression of myeloproliferative neoplasms.
