  • Exportin-5 is involved in microRNA precursor nuclear export. Dicer mRNA is now found to compete with pre-miRNAs for exportin-5 binding, suggesting that cytoplasmic levels of Dicer and its pre-miRNA substrate engage in a feedback loop. Analysis of adenoviral RNA indicates that it can also compete for exportin-5, suggesting that this might be a mechanism by which adenovirus can interfere with host miRNA processing.

  • Many Amyotrophic Lateral Sclerosis (ALS)-linked mutations cause accumulation of stress granules, and most ALS cases are caused by repeat expansions in C9ORF72. Here the authors show that C9ORF72 and the autophagy receptor p62 interact to associate with proteins symmetrically dimethylated on arginines such as FUS, to eliminate stress granules by autophagy.

  • RNA-based viruses can be engineered to express artificial microRNAs (amiRNAs). Here, the authors identify a candidate amiRNA that confers a replicative advantage to oncolytic viruses, enhancing their anticancer potency, and show that intercellular transfer of extracellular vesicles carrying the amiRNA promotes bystander killing of uninfected cancer cells.

  • Voinnet and colleagues show that autophagy targets RNAi components DICER and AGO2 for degradation when they are not bound to miRNA. The autophagy receptor NDP52 is required for this homeostatic regulation of the RNAi machinery. The authors also found that autophagy influences the post-translational regulation of DICER mRNA.

  • The ESCRT complex mediates sorting of ubiquitylated endosome-associated proteins into multivesicular bodies (MVBs). The RNA-induced silencing complex (RISC) components GW182 and AGO2 localize to membrane structures that congregate with MVBs. Loss of ESCRT function compromises miRNA-mediated silencing and increases GW182 levels, suggesting that ESCRT regulates RNAi by acting on GW182 turnover.
