Fu et al. uncover the features that allow the HIV capsid to cross the permeability barrier of nuclear pores in an importin-independent manner, explaining how the viral genome can reach the nuclear compartment when infecting nondividing cells.

Mechanical control of nuclear import by Importin-7 is regulated by its dominant cargo YAP

The translation of mechanical cues into gene expression changes is dependent on the nuclear import of mechanoresponsive transcriptional regulators. Here the authors identify that Importin-7 drives the nuclear import of one such regulator YAP while YAP then controls Importin-7 response to mechanical cues and restricts Importin-7 binding to other cargoes.

Recapitulation of selective nuclear import and export with a perfectly repeated 12mer GLFG peptide

The permeability barrier of nuclear pore complexes blocks passage of inert macromolecules but allows rapid, receptor-mediated, and RanGTPase-driven transport of cargoes up to ribosome size. The authors now show that such a barrier can be faithfully recapitulated by an ultimately simplified FG phase assembled solely from a tandemly repeated 12mer GLFG peptide.

How a variety of nuclear export signals (NESs) are recognized by their receptor CRM1 is now examined by crystallography, NMR spectroscopy, biochemistry and cellular approaches. The work reveals that the different NES peptides adopt different conformations to fit into five rigid hydrophobic binding pockets on CRM1, and redefines an NES consensus.
