Characterising acute and chronic care needs: insights from the Global Burden of Disease Study 2019

Chronic care manages long-term, progressive conditions, while acute care handles short-term ones. Here, authors show chronic conditions account for most of the global health burden, with 68% of DALYs and 93% of YLDs attributed to chronic care needs.

Molecular correlates of cisplatin-based chemotherapy response in muscle invasive bladder cancer by integrated multi-omics analysis

There are currently only a few biomarkers to predict the response of muscle invasive bladder cancer to therapy. Here, the authors analyse 300 tumors using exome and RNA sequencing and find that tumors with a high degree of genomic instability and a non-basal/squamous gene expression subtype are most likely to respond to treatment.

Discovery of rare variants associated with blood pressure regulation through meta-analysis of 1.3 million individuals

Meta-analyses in up to 1.3 million individuals identify 87 rare-variant associations with blood pressure traits. On average, rare variants exhibit effects ~8 times larger than the mean effects of common variants and implicate candidate causal genes at associated regions.

Re-analysis of public genetic data reveals a rare X-chromosomal variant associated with type 2 diabetes

Genome-wide association studies have uncovered several loci associated with diabetes risk. Here, the authors reanalyse public type 2 diabetes GWAS data to fine map 50 known loci and identify seven new ones, including one near ATGR2 on the X-chromosome that doubles the risk of diabetes in men.

Subgrouping patients with ischemic heart disease by means of the Markov cluster algorithm

Haue, Holm et al. use unsupervised clustering to subgroup a large set of ischemic heart disease patients from their pre-existing diseases, covering the entire multimorbidity spectrum. They identify subgroups that have different risk profiles and are characterized by different patterns when looking at patient blood tests and genetic profiles.

They describe current knowledge of tumour evolution, genomic heterogeneity and different tumour subtypes, as well as morphological heterogeneity seen in variant bladder cancer histology.
